I would spell it Charlie or Charley (the only really intelligent-looking spellings to me), and I would use it as a nickname for Charlaine. Charlotte has so many prettier nicknames that I wouldn't ever use Charlie as a short form for it.
Charlie on its own isn't at all strong enough, IMO. I love masculine nicknames for girls (Billie, Bobbie, Jimmie, Johnnie, Charlie, Sid, Mack), but I don't like them unless they've got a feminine full name to fall back on.
